If my truck has a one piece hub and rotor assembly will this replace that whole assembly? I know I would have to get rotors, but its unclear if my 99 f150 4.6 2wd has the one piece assembly or not.

do i need any specail tools or extra grease to pack bearings ? or does it just pop off the old and put on the new ?

You dont need a puller but some times it helps. It axle it designed to slide in and out of the center of the hub. If it gets a little sticky use PB blaster and tap the axle inward with a rubber mallet or a peice of wood and a hammer, that should brake it loose and you shold have no issues.
